(5) CD4 Single-Platform.

The most modern flow cytometers dedicated to clinical absolute counting, have been designed to operate as a single unit on a  'single-platform'. 

This avoids the problems of co-ordinating the matching populations between the two machines (reviewed in [6]) .
These 'single-platforms' are of  two major types:
Volumetric flow cytometers (a) determine the number of cells in a given volume of sample and directly provide the concentration of cells identified by various MAbs. These counters include the ORTHO Cytoron-absolute and the DAKO Galaxy (plus its manufacturer's version referred to as PARTEC PAS)
Precision-made microspheres, or (micro)beads (TruCount® or FlowCount®), can be added in known concentration to the tubes to count absolute numbers on flow cytometers that are not equipped with volumetric facility (b).

Both volumetric and bead-based methods yield accurate absolute counts.  

The excellent agreements between the volumetric and 'bead-based' single-platforms are shown using Bland-Altman statistics in a Figure taken from [11] (c). See also  [7].
